Bloo

25、d collection and processing are performed in accordance with DIN 58905-1. Im Gegensatz zu vielen anderen Gerinnungs-proteinen ist der VWF relativ stabil. Transport von Plasma oder Blut bei Raumtemperatur (RT) fhrt zu keinerlei Vernderungen, solange 48 h nicht ber-schritten werden. Danach kommt es zu

26、 einem graduellen Verlust groer Multimere und zu einem dysproportionalen Abfall funktioneller Teste wie VWF:RCo und VWF:CB, whrend das VWF:Ag ber noch lngere Zeit stabil bleibt 1. Hmo-lytische Proben knnen ein Hinweis auf fehlerhafte Blutentnahme und Lagerung sein und sind nicht zu verwenden. Die Tr

27、ansport- und Lagerungsbedin-gungen sind der Tabelle 1 zu entnehmen. In contrast to many other coagulation proteins, the VWF is relatively stable. The transport of plasma or blood at room temperature (RT) does not lead to any modifications, as long as a duration of 48 h is not exceeded. Afterwards a

28、gradual loss of large multimeres and a dysproportionate reduction in functional tests, such as VWF:RCo and VWF:CBassays, will occur while the VWF:Ag remains stable for a longer period of time 1. Haemolytic samples can be an indication of improper blood collection and storage and shall not be used. T

34、ided); drying oven for the incubation of ELISA plates.7.2 Materialien und Reagenzien 7.2 Materials and reagents 7.2.1 ELISA-Platten Als ELISA-Platten stehen Platten mit 96 Vertie-fungen (96 well) zur Verfgung, die eine Beschich-tung aufgrund physikalischer Adsorption erlauben und vorzugsweise aus Po

35、lystyrol hergestellt sind. Es sind Plattentypen mit hoher Proteinbindungs-kapazitt zu bevorzugen. Alternativ dazu ist auch die kovalente Immobilisierung von Typ III Collagen aus humaner Plazenta an praktivierte ELISA-Platten ber einen Spacer-Arm beschrieben 2. Bei Verwendung einer neuen Charge von P

36、latten ist es empfehlenswert, die Eignung in Kombination mit dem jeweiligen Collagen zu testen. Darber hinaus ist exemplarisch die Homogenitt der Beschichtung in Sonderheit in den Randbereichen der Platte nachzuweisen. 7.2.1 ELISA plates The available ELISA plates are plates with 96 wells, where coa

37、ting is possible as a result of physical adsorption and which are preferably made of polystyrol. Plate types having a high protein binding capacity shall be given preference. Alternatively, the covalent immobilization of type III collagen from human placentas onto pre-activated ELISA plates via a sp

38、acer arm is described 2. When using a new batch of plates, it is to be recommended to test their suitability in combination with the relevant collagen. In addition, the coating homogeneity, particularly that in the boundary areas of the plates, shall be demonstrated exemplarily. 7.2.2 Collagen Als g

39、eeignete Collagene haben sich kommerziell erhltliche Prparationen der Typen I (z. B. fibrillres Collagen aus der Pferdesehne) oder Typ III (z. B. Collagen aus humaner Plazenta) erwiesen 3. Da diese im Handel erhltlichen Collagene normalerweise nicht speziell fr die Durchfhrung des Collagenbindungste

40、sts herge-stellt werden, ist jede Charge in Vorversuchen auf ihre Eignung zu prfen. Typische Beschichtungs-konzentrationen sind fr Typ I Collagen (fibrillr, Pferdesehne) (10 bis 50) g/ml, fr Typ III Collagen (humane Plazenta) (1 bis 5) g/ml. 7.2.2 Collagen Commercially available preparations of type

41、 I (e.g. fibrillar collagen from horse tendons) or type III(e.g. collagen from human placentas) have proven to be suitable collagens 3. Since, normally, these commercially available collagens are not specially made for performing the collagen binding assay, each batch shall be tested for suitability

42、 in pre-tests. 43、Antikrper Hoch spezifische polyklonale anti-VWF Antikrper (beispielsweise aus dem Kaninchen) sind kommer-ziell erhltlich und knnen als Enzymkonjugate (z. B. als Peroxidase-Konjugat) verwendet werden.7.2.3 Antibodies Highly specific polyclonal anti-VWF antibodies (for example from rabbits) are commer

44、cially available and can be used as enzymatic conjugates (e.g. as peroxidase conjugate). 7.2.4 Puffersysteme Beschichtungslsungen: Fibrillre Collagene sind in der Regel nur im sauren Milieu lslich, fr die Beschichtung sind daher saure Lsungen (pH 4) zu empfehlen. Darber hinaus knnen auch hochionisch

45、e Lsungen unter Zusatz von stabilisierenden Kohlenhydraten ver-wendet werden. Die Beschichtung mit Typ-III-Collagenen kann gegebenenfalls auch bei neut-ralen pH-Werten, beispielsweise in PBS-Puffer, pH 7,3, durchgefhrt werden. 7.2.4 Buffer systems Coating solutions: As a rule, fibrillar collagens ar

46、e soluble in an acidic environment only; therefore, acidic solutions (pH value 4) are to be recommended for the coating. Furthermore, highly ionic solutions can be used to which stabilizing carbohydrates have been added. If applicable, coating with type III collagens can also be carried out at neutral pH values, for example in a PBS buffer w
