21、tes erfolgen nach DIN 58905-1. Blood collection and processing are performed in accordance with DIN 58905-1. Im Gegensatz zu vielen anderen Gerinnungs-proteinen ist der VWF relativ stabil. Transport von Plasma oder Blut bei Raumtemperatur (RT) fhrt zu keinerlei Vernderungen, solange 48 h nicht bersc

22、hritten werden. Danach kommt es zu einem graduellen Verlust groer Multimere und zu einem dysproportionalen Abfall funktioneller Teste, whrend das VWF:Ag ber noch lngere Zeit stabil bleibt 1. Hmolytische Proben knnen ein Hinweis auf fehlerhafte Blutentnahme und Lagerung sein. Die Transport- und Lager

28、.1 Gerte, Materialien und Reagenzien Der Ristocetin-Cofaktor Test wird bei Anwendung der makroskopischen Variante sowie unter Verwen-dung von Aggregometern oder Gerinnungsanaly-sensystem in der Regel mit kommerziell erhlt-lichen Testvarianten durchgefhrt. Bei Durch-fhrung des Tests mit Hilfe des mak

29、roskopischen Agglutinationstests wird eine Glasplatte bentigt. Bei Durchfhrung des Tests mit Hilfe eines klassischen Aggregometers wird ein Instrument zur photometrischen Bestimmung der Thrombozyten-aggregation mit instrumentspezifischen Kvetten, Rhrern oder Magneten zur Durchfhrung der Bestimmung b

30、entigt. Auerdem sind Pipetten und Mixer notwendig. Darber hinaus kann der Test auch an einigen vollautomatischen Gerinnungs-analysensystemen mit Hilfe einer turbidimetrischen Bestimmung der Thrombozytenagglutination durch-gefhrt werden. 6.1 Apparatus, materials and reagents When applying the macrosc

31、opic variant and using aggregometers or coagulation analysis systems, the ristocetin cofactor assay is usually performed employing commercially available test variants. When carrying out the test by means of the macroscopic agglutination assay, a glass plate is necessary. When performing the test us

32、ing a classic aggregometer, a photometric thrombocyte aggregation determination instrument comprising the instrument-specific cells, stirrers or magnets is required to carry out the determination. In addition, pipettes and mixers are needed. Furthermore, with some fully automated coagulation analysi

33、s sys-tems, the assay can be performed by means of a turbidimetric determination of the thrombocyte agglutination. 6.1.1 Thrombozytensuspension Zur quantitativen Bestimmung des Ristocetin-Cofaktors knnen standardisierte Thrombozyten-prparationen verwendet werden, die durch Zentri-fugation aus antiko

34、aguliertem Vollblut oder mit Hilfe anderer maschineller Verfahren gewonnen werden. Die Thrombozyten zur Bestimmung des Ristocetin-Cofaktors werden in fixierter Form (Formaldehyd, Paraformaldehyd) eingesetzt. Zur Verbesserung der Lagerstabilitt der Thrombozyten ist Lyophili-sieren optional mglich. 6.

35、1.1 Thrombocyte suspension For the quantitative determination of the ristocetin cofactor, standardized thrombocyte preparations can be used which are obtained by centrifugation from anticoagulated whole blood, or by other automatic procedures. The thrombocytes for the determination of the ristocetin

36、 cofactor are used in a fixed form (formaldehyde, paraformaldehyde). In order to improve the storage stability of the throm-bocytes, optional lyophilization is possible. Therefore, the constancy of, and the compliance with, the required performance data in the ristocetin cofactor assay shall be tested for each new batch used (see 6.3). 6.1.3 Referenzplasma Das Normalplasma muss (1,00 0,20) IU/ml (100 20) % der Norm) VWF
