Rhrchen DIN 58970 14 100 G1,mindestens 5 min auf eine Temperatur von (37 t 0,5)9Cvorgewrmt. Anschlieend wird das vom Hersteller vor-geschriebene Volumen Th

30、romboplastinsuspension, dienach vorschriftsmiger Rekonstitution im Wasserbad auf(37 t 0,5)9C vorgewrmt wurde, mit einer Kolbenhub-pipette nach DIN 12650-2 einpipettiert. Das exakt abge-messene Kapillarblutvolumen wird unmittelbar nach derEntnahme in die vorbereitete Gewebethromboplastin-suspension1)

31、ausgeblasen und durch leichtes Schttelngemischt. Beim Ausblasen soll die Pipettenspitze dichtber der Oberflche der Reagenzlsung an der Innen-wand des Glasrhrchens anliegen.Gleichzeitig mit der Zugabe der Suspension wird eineStoppuhr in Gang gesetzt. Ein Hkchen aus Platin wird inregelmigen Abstnden v

32、on etwa 0,5 s durch dasReaktionsgemisch gefhrt bzw. das Glasrhrchen in Zeit-abstnden von etwa 1 s vorsichtig gekippt. 35、T DIN 58910 D5 Blood collectionA cleaned and dry spot on the fingertip or earlobe of thepatient shall be punctured by means of a sterilesingle-use lancet. The cut shall be deep enough to allowspontaneous blood flow. Pressing and squeezing is notpermissible.The first issuing blood drops shall be used

36、 for the deter-mination. The specified quantity of blood (10 ml to 50 ml)shall be drawn within 10 s by means of a sterilenonactivating single-use pipette or capillary tube byholding the tip of the pipette or capillary tube in ahorizontal position into the issuing blood drops.The determination of the

37、 thromboplastin (prothrombin)time shall be carried out immediately afterwards.6 ProcedureThe manual hook technique or tilt-technique shall beused to perform the determination.Unsiliconized centrifuge glass tubes, e.g. tubesDIN 58970 14 100 G1, are prewarmed for at least5 min in a waterbath to a temp

38、erature of (37 t 0,5)9C. Thevolume of thromboplastin suspension specified by themanufacturer is reconstituted according to the instruc-tions and prewarmed to (37 t 0,5)9C in a waterbath. Itshall be pipetted into the tubes by means of a pistonpipette as described in DIN 12650-2. The exactlymeasured v

39、olume of capillary blood shall be blown off intothetissuethromboplastinsuspension1)immediately afterblood collection and mixed by gently tilting it back andforth. When blowing it off, the tip of the pipette shall beclose to the surface of the reagent solution and in contactwith the inner wall of the

40、 glass tube.A stopwatch is started simultaneously when adding thesuspension. A platinum hook is passed through thereaction mixture at regular intervals of about 0,5 s or theglass tube shall be gently tilted back and forth at intervalsof 1 s. The stopwatch is stopped when the first clot can bedetecte
