Fr ein Monitoring von GVOwird daher ein Verfahren bentigt, das die Erfassungund Dokumentation von Eintrag und Verbreitung vonGVO be

19、r Pollen in einem bundesweit und natur-raum-reprsentativ angelegten Messnetz ermglicht.Hierzu werden biologische und technische Pollen-sammler (VDI 4330 Blatt 3) sowie PCR-Screening-Verfahren zum Nachweis des GVO-Eintrages(VDI 4330 Blatt 7) eingesetzt. In der vorliegendenRichtlinie wird ein biologis

20、ches Sammelsystem be-schrieben.IntroductionPollen dispersal plays a significant role in the spreadof genetically modified organisms (GMO). Hence, amonitoring procedure that involves recording anddocumentation of input and distribution of GMO viapollen in a national measurement network mirroringthe n

21、atural environment is required. For this, techni-cal and biological samplers of pollen (VDI 4330Part 3) as well as PCR-screening procedures are em-ployed to proof GMO-input (VDI 4330 Part 7). 23、eren Erfassung des inder Luft verteilten Pollens korreliert hoch mit dervorherrschenden Windrichtung sowie der Auspr-gung der Landschaft durch die Hhe des Bewuchsesmit Pflanzen.Bienenvlker und hier insbesondere die sammeln-den Flugbienen durchstreifen aktiv einen Raum undsind daher flchenbezogene Sa

24、mmler. Die Samme-laktivitt und rumliche Nutzung ist primr abhngigvon dem zur Verfgung stehenden Nektar- und Pol-lenangebot (= Trachtangebot) in der Raumzelle. EinBienenvolk befliegt in der Regel ein Areal mit einemRadius von 3 km.1 AnwendungsbereichDiese Richtlinie beschreibt ein Verfahren, mit demP

25、ollen insbesondere auch Pollen von gentechnischvernderten Organismen mittels Bienenvlkern ge-sammelt werden knnen. Die aktiven Sammelbienensammeln den Pollen, tragen ihn in das Bienenvolkund speichern ihn dort in Pollenzellen (Bienenbrot).Weiterhin tragen Sammelbienen Nektar und Honig-tau ein. Letzt

26、genannte Sammelgter enthalten Pollen,der aus den Antheren der Blte in den Nektartropfengefallen ist oder Pollen, der durch den Wind verbreitetim Nektar anderer Blten bzw. auf klebrigem Honig-tau an Pflanzen haften bleibt. Im Bienenvolk werdenNektar und Honigtau durch die Bienen zu Honig kon-vertiert

27、 und gelagert.Honig und Bienenbrot knnen als Proben fr die an-schlieenden Pollenanalysen genutzt werden, da esmglich ist, ausreichende Pollenmengen fr eine mo-lekularbiologische Diagnostik anzureichern.Notwendige Grundlagen zum Verstndnis dieserRichtlinie sind in den Richtlinien VDI 4330 Blatt 1und

28、VDI 4330 Blatt 2 dargelegt. Die Sammlung vonPollen in Honig und Bienenbrot ist in einem unmittel-baren Zusammenhang mit der technischen Sammlungfr das GVO-Monitoring zu sehen (VDI 4330 Blatt 3).Der Einsatz der biologischen, aktiven Sammlerin Ho-nigbiene und der technischen Passivsammler ergn-zen sic

29、h in vielfltiger und positiver Weise fr dasPollenmonitoring von GVO. Whrend die techni-schen Sammler Ergebnisse zum Polleneintrag amStandort liefern, zeigen die Bienen eine raumber-greifende Sammelaktivitt, die einen Querschnittber die im Raum etablierten blhenden Pflanzen ab-geben. Mit beiden Samme

30、lverfahren wird ein breitesPollenartenspektrum erfasst, wobei sich die Verfah-ren ber die Vegetationszeit ergnzen 1.It is reasonable to use both technical and biologicalsamplers of pollen. The technical samplers (VDI 4330Part 3) are stationary point-samplers. Their recordingof pollen dispersed in th

31、e air correlates with the pre-vailing wind direction and is characteristic of thelandscape according to the height of the natural veg-etation. Bee colonies, especially the foraging bees, activelyroam an area and are therefore area related sampler.Collecting activities and spatial utilisation is prim

32、a-rily dependent on the availability of nectar and pollensources (= supply of melliferous plants) within thespatial zone. A colony of bees normally forages overan area of 3 km radius.1ScopeThis guideline describes a procedure through whichpollen in particular pollen of genetically modifiedorganisms

33、may be sampled by means of bee colo-nies. Active foraging bees gather the pollen, carry itinto the bee colony and store it there in pollen cells(bee-bread). Furthermore, foragers gather nectar andhoneydew. The latter goods contain pollen which fellfrom the anthers of the blossom into the nectar drop

34、or pollen, which was dispersed by the wind and sticksin the nectar of other blossoms or adheres to thesticky honeydew of plants. Nectar and honeydew areconverted to honey and stored by the bees in the bee-hive. Honey and bee-bread may be used as samples for thesubsequent analysis of pollen as it is

35、possible to con-centrate sufficient amounts of pollen for a molecularbiological diagnostic.Guidelines VDI 4330 Part 1 and VDI 4330 Part 2present the necessary fundamentals for the under-standing of this guideline. The sampling of pollen inhoney and bee-bread shall be viewed in conjunctionwith the te

36、chnical sampling for the GMO-monitoring(VDI 4330 Part 3). The use of the biological, active foraging honeybeeand the technical passive samplers complement eachother in a multifaceted and positive way for pollenmonitoring of GMO. Whereas the technical samplersprovide results to pollen yields at the s

37、ite, bees dis-play a spatially overlapping sampling activity, whichrepresents a cross section of the established, blos-soming plants in the area. A wide spectrum of pollenspecies is recorded using both sampling methodswith the procedures complementing each otheracross the vegetation period 1.B55EB1B

Bienenvlker werden in dem zuuntersuchenden Areal positionie

39、rt (Verbringen vonBienenvlkern) oder bereits dort vorhandene, festlokalisierte Bienenvlker (Bienenvlker von Stand-imkern) werden genutzt.Durch den Eintrag von Nektar, Honigtau und Pollentragen die Bienen einen Teil der im Areal zu der Zeitvorhandenen Pollen ein. Im Honig und Bienenbrotsind diese Pol

40、len eingelagert und stehen fr sptereAnalysen zur Verfgung.Das von den Bienen genutzte Areal hngt von ver-schiedenen Faktoren ab (Wetter, Trachtangebot,Landschaftsnutzung und -struktur etc).Honig birgt gegenber Bienenbrot folgende be-deutende Vorteile: kontinuierlicher Eintrag, sichereVerfgbarkeit, w

41、esentlich bessere Matrix fr licht-mikroskopische und molekularbiologische Analysen. Die Expositionszeit kann flexibel von minimal fnfTagen bis zu mehreren Wochen festgelegt werden.Bei mehr als einer Woche Expositionszeit sind auchProbenahmen in Intervallen mglich. Das Probenmaterial Honig wird licht

42、mikroskopisch(Palynologie) und molekularbiologisch (PCR) unter-sucht.3 Probenahmeeinrichtung3.1 BienenvolkBienenvolk soll hier definiert sein als ein Bienenvolk(Wirtschaftsvolk der Honigbienenart Apis mellifera),gefhrt durch den Imker (w/m) nach guter imkerli-cher Praxis. Das Bienenvolk umfasst die

43、Beute (Kas-ten i.w.S. als Behausung fr die Bienen), Wabenbau,eine Knigin, 15.000 bis 40.000 Arbeitsbienen sowiein bestimmten Monaten mehrere hundert Drohnen.Mindestens ein Bienenvolk wird an dem festgelegtenStandort aufgestellt. Die exakte Aufstellung erfolgtnach guter imkerlicher Praxis (Sicherung

44、gegenberHochwasser, Sturm, Astschlag etc.) 2; 3. Eine re-gelmige Betreuung des Bienenvolkes (ca. alle 9 bis14 Tage) ist sicherzustellen. 3.1.1 BienenwohnungGre der Beute und Anzahl der Waben sind nach gu-ter imkerlicher Praxis der Gre des Bienenvolkessowie dem Angebot an Nektar, Honigtau und Pollena

Magazinbeuten sollten bevorzugt werden.Magazinbeuten sind durch ein Modulsystem sehr fle-xibel an die jeweiligen Erfordernisse anzupassen.2 Basics of procedureThe Bee colony serves as biological active sampler ofpollen. The bee colonies are positioned within thearea under investigation i.

46、e. relocated bee colonies orthose bee colonies are used which are already presenti.e. permanent apiaries by local beekeepers. By gathering nectar, honeydew and pollen, bees col-lect a fraction of the pollen present at the time in thearea. These pollen are stored in wax combs as honeyand bee-bread an

The area used by the bees depends on various factors(weather, availability of meltiferous plants, utilisa-tion of landscape and landscape structure, etc.).Honey has significant advantages over bee-bread:continuous yield, guaranteed availability and a sig-nificant

48、ly better matrix for light microscopic andmolecular biological analyses.Exposure time may be flexibly specified from a min-imum of five days up to several weeks. For exposuretimes of more than a week, sampling in intervals isalso possible. The sampling material honey is analysed using lightmicroscop
