9、reagents for microbiological tests. 3 Equipment Use equipment that is as specified in SANS 171. 4 Media and reagents 4.1 General Use ingredients for media and reagents of the quality specified in SANS 5553, and use the procedures specified in that method. 4.2 Nutrient agar Ingredients: agar 15,0 g b
10、eef extract 1,0 g peptone 5,0 g sodium chloride. 5,0 g yeast extract 2,0 g Dissolve the ingredients in water and dilute to 1 L. So adjust the pH value that, after sterilization, it will be 7,1. NOTE Additional organisms ma
12、y be used if so desired. 5.2 Preparation of test organism suspensions Maintain the test organisms and prepare the cultures for test suspensions as described in SANS 636. Using sterile water as the diluent, prepare suspensions as described in SANS 636, but containing (10 4 10) % living cells per mill
13、ilitre. 6 Test procedure 6.1 Using a sterile pipette, transfer 10 mL of the test sample to each of four sterile bottles having a nominal capacity of 30 mL and which are fitted with screw closures. 6.2 Transfer 0,1 mL of S. aureus test organism suspension (see 5.2) to each of the four bottles and mix
14、 well, avoiding the formation of air bubbles. Allow the bottles to stand at ambient temperature. 6.3 One hour after adding the test organism suspension, take two 1 mL volumes from one of the bottles and transfer each volume to a plate (Petri dish). For each separate plate take one 15 mL volume of nu
15、trient agar (see 4.2) which has been melted and tempered to 45 C, add it to the plate, and mix well to ensure an even distribution of colonies after incubation. Avoid spilling any of the contents of the plate during this process. Allow the agar to set, invert the plates, transfer them (suitably labe
16、lled) to an incubator, and incubate at 37 C. 6.4 After 48 h incubation, remove the plates from the incubator and count the colonies which have developed. Record these results. 6.5 Calculate the total count of S. aureus by averaging the counts on each plate. 6.6 Repeat the procedure in 6.3 to 6.5 (in
17、clusive) for each of the three remaining bottles after they have been allowed to stand at ambient temperature for 24 h, 7 d, and 28 d respectively. 6.7 Carry out a control test by transferring 10 mL of sterile water to a further screw-capped bottle. Transfer 0,1 mL of S. aureus test organism suspens
18、ion (see 5.2) to the bottle, mix well and immediately take two 1 mL volumes and transfer each volume to a plate (Petri dish). Pour the plates, mix, invert, incubate, count, and record the colonies developed as in 6.3 and 6.4. 6.8 Calculate the total control count of S. aureus as in 6.5. 6.9 Repeat t
